Evidence Tier: FDA-Approved — Tirzepatide is a first-in-class dual glucose-dependent insulinotropic polypeptide (GIP) and GLP-1 receptor agonist. Developed by Eli Lilly, it represents the next generation of incretin-based therapies.
Overview
Tirzepatide (Mounjaro for T2D, Zepbound for obesity) is a once-weekly injectable peptide that simultaneously activates both the GIP and GLP-1 receptors. This dual mechanism produces effects on blood glucose and body weight that exceed those of GLP-1 receptor agonists alone.
Key Clinical Data
- SURPASS Trials: HbA1c reduction up to 2.4% vs placebo at 26 weeks
- SURMOUNT Trials: Weight loss of 15-20% at 72 weeks (SURMOUNT-1, n>7,700)
- Superior to semaglutide in head-to-head trials for both glucose control and weight loss
