KPV (Lysine-Proline-Valine)

Overview

KPV (Lys-Pro-Val) is the C-terminal tripeptide of alpha-melanocyte-stimulating hormone (alpha-MSH). Unlike the full alpha-MSH hormone, KPV retains anti-inflammatory activity without melanogenic (tanning) effects. It has been studied primarily in models of intestinal inflammation and immune disorders.

Mechanism of Action

  • NF-kB Inhibition: Directly enters cells and inhibits NF-kB nuclear translocation, reducing inflammatory gene expression
  • Cytokine Suppression: Reduces IL-1beta, IL-6, TNF-alpha production in immune cells
  • Gut Barrier Enhancement: Improves intestinal epithelial barrier function in colitis models

Regulatory Status

FDA: Scheduled for PCAC evaluation July 23-24, 2026.
